Приступив в конце 1980 г. к созданию самолета А300-600, консорциум в его конструкции использовал переднюю и центральную секции фюзеляжа и крыло самолетов А300В2/В4, а хвостовую часть фюзеляжа от самолета А310. Кабина экипажа такая же, как и на самолете A310. С целью обеспечения требуемой центровки фюзеляж удлинен на 0,53 м, на концах крыла установлены стреловидные "шайбы". Первый опытный самолет (с двигателями JT9D-7R4H1) выполнил первый полет 8 июля 1983 г. В начале марта 1984 г. он был сертифицирован во Франции, а в конце марта первый самолет был поставлен авиакомпании "Сауди Эрлайнз". В марте 1985 г. была завершена сертификация варианта самолета с двигателями CF6-80C2. В сентябре начались его поставки; первой его получила таиландская авиакомпания "Тай Эруэйз". На основе самолета А300-600 в 1988 г. был разработан вариант с увеличенной дальностью полета A300-600R. Самолет оснащен цифровыми комплексами авионики EFIS и ЕСАМ, которые используются на самолетах А310. Выпускался серийно в 1984 -1991 гг. Всего было поставлено 35 самолетов данной модификации, к началу 1996 г. в эксплуатации находилось 35. Летно-технические характеристики
